I've done it just the usual way: Outlines --> Colored and then used the marker for the structures --> Just added some effects like Luminosity/Addition, Screen, Overlay and Multiply for the needed areas and Voilà OwO'

It's harder to describe than to show q~q'

I'm bad at painting and my English is also horrible x3
Sometimes I use the video transmitting function of Skype to show my friends something ^^°
